ADP‐heptose enables Helicobacter pylori to exploit macrophages as a survival niche by suppressing antigen‐presenting HLA‐II expression

چکیده
The persistence of Helicobacter pylori in the human gastric mucosa implies that immune response fails to clear infection. We found H. compromises antigen presentation ability macrophages, because decline presenting molecules HLA-II. Here, we reveal main bacterial factor responsible for this effect is ADP-heptose, an intermediate metabolite biosynthetic pathway lipopolysaccharide (LPS) elicits a pro-inflammatory epithelial cells. In it upregulates expression miR146b which, turn, would downmodulate CIITA, master regulator HLA-II genes. Hence, pylori, utilizing exploits specific arm macrophage establish its survival niche face defense elicited mucosa.

Chlamydia exploit the mammalian tryptophan-depletion defense strategy as a counter-defensive cue to trigger a survival state of persistence
We previously proposed that in Chlamydiaceae rapid vegetative growth and a quiescent state of survival (persistence) depend upon alternative protein translational profiles dictated by host tryptophan (Trp) availability. These alternative profiles correspond, respectively, with a set of chlamydial proteins having higher-than-predicted contents of Trp ("Up-Trp" selection), or with another set exh...

متن کاملmicroRNA-146a promotes mycobacterial survival in macrophages through suppressing nitric oxide production
Macrophages play a crucial role in host innate anti-mycobacterial defense, which is tightly regulated by multiple factors, including microRNAs. Our previous study showed that a panel of microRNAs was markedly up-regulated in macrophages upon mycobacterial infection. Here, we investigated the biological function of miR-146a during mycobacterial infection. miR-146a expression was induced both in ...
